Tip: Make a party with two of the required characters + a fast group damage dealer and you can clear the character usage missions while also farming the required monsters. Some times, the required monsters will also be featured in the upcoming stages or lost chapters for easier completion.
Mindflayers + Skeletons
Note: You can complete both missions simultaneously using this option. For easier stages to farm each monster type individually, check the next section.
Story Act 2 Chapter 4 -> Abandoned Service Road Pt. 1 (Normal) has 5 Skeletons and 3 Mind Flayers in 4 waves, completing both missions in 6 fights.
Alternative:
Vayne’s Lost Chapter: Conqueror’s Duty Pt. 11 (Normal) has 3 Mind Flayers and 2 Skeletons in 3 waves, completing both missions in 10 fights.
Mindflayer x15
Story Act 2 Chapter 4 -> Uncanny Closet Pt. 1 (Normal) has 5 Mind Flayers in 4 waves, but is buried at the end of the chapter.
Skeleton x30
Story Act 2 Chapter 6 -> Small Hill Pt. 1 (Normal) has 8 Skeletons in 4 waves.
Alternative:
Ace’s Lost Chapter: Unparalleled Power Pt. 8 (Normal) has 6 Skeletons in 4 waves.
